The LAL reagent prepared from amebocyte lysate of the American horseshoe crabs is coagulated (gelated) by (1→3)-β-D-glucan in the specimen. This gelation reaction is captured as a change in transmitted light intensity, and the time from the start of the reaction to the point where a fixed transmitted light intensity ratio is reached (gelation time) is analyzed to calculate the concentration of (1→3)-β-D-glucan in the specimen.
